Stem Cell Storage Temperatures
1. Cryopreserved (Long-Term Use) Temperature: –150°C to –196°C
Storage Method: Liquid nitrogen (LN₂) vapor phase or liquid phase
Why: At this temperature, all metabolic activity stops, and cells can be preserved for years to decades without loss of function
Storing stem cells at warmer temperatures—even briefly—can reduce viability and compromise cell membrane integrity.
2. Short-Term Holding (Before Use or Shipping) Temperature: –80°C
Storage Method: Ultra-low mechanical freezer Why: Acceptable for short-term storage (days to a few weeks) for prepared therapeutic doses or ready-to-thaw clinical vials
Stem Nova Network, for example, ships stem cells frozen on dry ice (approx. –78.5°C) for transit, with the expectation that they are transferred to –80°C or colder immediately upon receipt.

Thawing Cells: Step-by-Step Procedure
1. Preparation of Warm Water
Warm sterile water in a 37°C water bath. Ensure that the water bath is calibrated correctly to maintain this temperature throughout the thawing process.
2. Surface Sterilization
Before handling the cell vial, wipe the outside with 70% ethanol or isopropanol to minimize contamination risks. This step is crucial for maintaining aseptic conditions.
3. Pressure Relief in Vial
In a biosafety cabinet, carefully twist the cap of the vial a quarter-turn to relieve any internal pressure that may have built up during freezing. Retighten the cap to prevent contamination.
4. Thawing Cells
- Place the vial in the 37°C water bath and gently shake it to facilitate thawing. Do not submerge the vial completely; only the lower part should be in contact with the warm water.
Monitor closely and remove the vial when only a small frozen cell pellet remains visible
(Thaw for NOT LONGER than 1 minute – to ice ball stage). Handle gently and completely avoid vortexing or shaking vigorously, as this will likely damage cells.
5. Second Surface Sterilization
Again, wipe the outside of the vial with 70% ethanol or isopropanol after thawing to ensure that any potential contaminants are removed before opening.
6. Dilution of Cells
Dilute the thawed cells by adding an equal volume of warmed culture medium (1:10 dilution). This step helps to reduce cellular stress and promotes recovery.
7. Centrifugation
Centrifuge the diluted cell suspension at 1200 RPM for 5 minutes max at room temperature (15 - 25°C). This process helps to pellet the cells while removing any cryoprotectant (e.g., DMSO) that may be present in the medium.
8. Supernatant Removal
After centrifugation, carefully remove the supernatant using a pipette. Leave a small amount of medium above the cell pellet to avoid disturbing it during resuspension.
9. Resuspension of Cell Pellet
Gently resuspend the cell pellet by flicking the tube lightly with your finger or using a pipette tip to mix very gently; Add 1 ml of solution; wait 1 min, slowly add another 2 ml of solution. Avoid vigorous mixing or vortexing, which could damage cells.
10. Final Medium Addition
After waiting another minute; slowly add up to 10 ml of the remaining warmed medium to the tube containing the resuspended cells. This gradual addition helpsto acclimate the cells back into their growth environment.
